Laser scanning confocal microscopy

Leica Stellaris 5 

  • 1 white light laser + 1 UV laser (405 nm), 200 excitation lines between 485 and 685 nm + 1 in the UV spectrum (405 nm): it can simultaneously excite different fluorochromes with 8 wavelengths;
  • Acousto-Optical Beam Splitter (AOBS);
  • 3 spectral Power HyD S detectors with high photon detection efficiency: compared to standard confocal microscopes, they increase the signal-to-noise ratio and the analytical detail of dynamic biological phenomena by 20 times, while decreasing the laser power required for fluorochrome excitation by 100 times, thereby reducing photobleaching and damage to live biological samples;
  • TauSense, a photon-counting technology that discriminates between different fluorochromes: it enables precise spectral separation of fluorescence emissions, excluding autofluorescence and reflection phenomena;
  • Spatial ultra-resolution (120 nm), twice that of standard confocal microscopes (240 nm).

Leica TCS-SP5

  • 1 Argon laser + 2 He/Ne lasers, 6 excitation lines in the visible spectrum;
  • Acousto-Optical Beam Splitter (AOBS), which allows the excitation light to be filtered with high precision;
  • 3 high-sensitivity spectral photomultiplier tube (PMT) detectors;
  • 1 detector for brightfield (DIC) acquisitions.
